Vy\u0159azen\u00ed proteasomu z\u00a0\u010dinnosti jeho zablokov\u00e1n\u00edm je velmi slibn\u00fd zp\u016fsob, jak postupovat proti n\u00e1dorov\u00e9mu bujen\u00ed bun\u011bk nebo mnoha\u010detn\u00e9ho myelomu. Vzorov\u00e9 slou\u010deniny jsou v sou\u010dasnosti v\u00a0z\u00e1v\u011bre\u010dn\u00e9 f\u00e1zi klinick\u00fdch test\u016f nebo ji\u017e byly FDA schv\u00e1leny pro klinick\u00e9 vyu\u017eit\u00ed. S l\u00e1tkami z na\u0161\u00ed laborato\u0159e sd\u00edlej\u00ed velmi podobn\u00e9 strukturn\u00ed motivy. Z toho d\u016fvodu se o\u010dek\u00e1vaj\u00ed stejn\u00e9 nebo podobn\u00e9 \u00fa\u010dinky. P\u0159ipraven\u00e9 slou\u010deniny budou otestov\u00e1ny jako inhibitory proteasomu, protein kin\u00e1z a t\u00e9\u017e jako slou\u010deniny schopn\u00e9 zp\u016fsobovat apopt\u00f3zu.<\/p>\n<p style=\"text-align: justify;\">Pro toto t\u00e9ma hled\u00e1me jak studenty\/-tky bakal\u00e1\u0159sk\u00fdch, tak magistersk\u00fdch program\u016f. V laborato\u0159i za\u010dnou p\u0159\u00edpravou, \u010di\u0161t\u011bn\u00edm a identifikac\u00ed z\u00e1kladn\u00edch intermedi\u00e1t\u016f, aby se pozd\u011bji mohli v\u011bnovat spojov\u00e1n\u00ed t\u011bchto z\u00e1kladn\u00edch stavebn\u00edch kamen\u016f v pokro\u010dilej\u0161\u00ed intermedi\u00e1ty nebo c\u00edlov\u00e9 molekuly.<\/p>\n<p style=\"text-align: justify;\"><strong>EN:<\/strong><\/p>\n<p style=\"text-align: justify;\">This basic research is focused to prepare new compounds (salicylamide \u2013 peptide aldehydes, various epoxides or boronic acids) which should join a large group of proteasome inhibitors. Inhibition of proteasome via blocking its protein recycling function is one of a very promising ways to treat tumor cells or multiple myeloma. Due to a very similar constitution of novel compounds which are now in last stage of clinical trials it can be expected very similar properties as well as biological activities. Prepared compounds are going to be tested as proteasomal and protein kinase inhibitors, for the type of caused apoptosis.<\/p>\n<p style=\"text-align: justify;\">For this research, we are looking for the bachelor as well as master students. They work will start with preparation of basic intermediates, their purification and identification.
